Pharmacology and Function of Tachykinin Receptors

چکیده
The existence of SP was described in the 1930s by Von Euler and Gaddum in mammals, and tachykinin peptides were identified and sequenced by Erspamer in non-vertebrated (eledoisin) and then in nonmammalian species (physalaemin). In the 1960s Lembeck continued Von Euler and Gaddum’s studies, describing the effects of SP and other tachykinins in the central nervous system (CNS) and peripheral preparations of mammals. The CNS physiology of SP was further developed by Konishi and Otsuka in the 1970s, whereas Hokfelt gave a major contribution in defining the neuroanatomical localisation of this peptide. Meanwhile, Chang and Leeman had defined the amino acid sequence of SP, which was then followed in the 1980s by the discovery of two other mammalian tachykinins, namely neurokinin A (NKA), also known as substance K, and neurokinin B (NKB), also known as neuromedin K, by Matsuo, Munekata and co-workers (Table 1). 2

The role of Tachykinins in health and disease
Background & Aim: Tachykinin peptide family is one of the largest peptide families in animals that has been first discovered in the gastrointestinal tract about 70 years ago and isolated for the first time from the small intestine and brain. Up to now, more than 40 tachykinins have been extracted from invertebrates (insects, worms and fungi) and vertebrates (skin, gastro-intestinal duct, centra...
متن کاملFunctional selectivity of NK1 receptor signaling: peptide agonists can preferentially produce receptor activation or desensitization.
A cascade of events follows binding of an agonist ligand to the tachykinin NK(1) receptor. These events include activation of multiple signal transduction pathways as well as cellular modulation of receptor function by the process of desensitization. This study examines the differences in the abilities of naturally occurring peptide agonist ligands of the tachykinin NK(1) receptor to preferenti...
متن کاملTachykinin family genes and their receptors are differentially expressed in the hypothyroid ovary and pituitary.
Plasma tachykinin levels are known to be altered with sexual acyclicity and loss of reproductive function. Ovulatory dysfunction, as seen in postmenopausal women, is also often encountered in hypothyroid patients. To know the involvement of different tachykinin genes in hypothyroidism-associated reproductive disorders, we performed DD-PCR with the pituitary RNA of control and hypothyroid rats t...
متن کاملTachykinin receptor expression and function in human esophageal smooth muscle.
Tachykinins are present in enteric nerves of the gastrointestinal tract and cause contraction of esophageal smooth muscle; however, the mechanisms involved are not understood. Our aim was to characterize tachykinin signaling in human esophageal smooth muscle. We investigated functional effects of tachykinins on human esophageal smooth muscle using tension recordings and isolated cells, receptor...
متن کاملNeurokinin A-induced vasoconstriction and muscular contraction in the rat isolated stomach: mediation by distinct and unusual neurokinin2 receptors.
This study examined the pharmacological identity of the tachykinin receptors which in the rat stomach mediate vasoconstriction and muscular contraction. The vasculature of the rat isolated stomach was perfused with oxygenated Krebs buffer containing 3% dextran. Vasoconstrictor responses were recorded as increases in the vascular perfusion pressure and gastric contractions were measured as incre...

The biochemical and pharmacological properties of a novel antagonist of the tachykinin neurokinin 1 (NK1) receptor, SSR240600 [(R)-2-(1-[2-[4-[2-[3,5-bis(trifluoromethyl)phenyl]acetyl]-2-(3,4-dichlorophenyl)-2-morpholinyl]ethyl]-4-piperidinyl)-2-methylpropanamide], were evaluated. SSR240600 inhibited the binding of radioactive substance P to tachykinin NK1 receptors in human lymphoblastic IM9 c...
